Radioisotopes are used in SPECT scans. The scan is then able to track blood flow to certain areas of the brain by tracking the distribution of these radioisotopes around the brain. A higher amount of radioisotopes in a given area of the brain = greater blood flow to that area = greater activity in that area.

A PET scan uses radioactive glucose, but a SPECT scan doesn't :)
