Question on how rounds work

Ok, so I got a scholarship from Monash and I have already accepted it.
Since Monash was my first preference, if I accepted their course offer in the first round, would that mean that my second preference (RMIT) would not make me an offer in the second round?

Yes, since you got your Preference #1 in Round 1, there’s no other higher preference to offer (assuming you don’t move any preferences).

The only exception is if you decide to move your RMIT engineering course to first preference between rounds and your Monash engineering course into a lower preference. In this latter case, if you are offered the RMIT engineering course in subsequent offer rounds, you might lose the Monash scholarship (many scholarships have the condition that you must be enrolled in that institute’s course to have the scholarship).
