Ultimately, you want a more fluid plasma membrane. And what can turn things to mush? Heat and electricity!!!!! This is achieved by what you have mentioned, electroporation!!!, where an electric current is administered to the bacterial cell. It is this electric current that alters the plasma membrane, increasing its permeability so the plasmid can scoot through. Likewise, another method known as heat shock can be used as well. This uses knowledge of how temperature can affect the fluidity of the membrane. The bacteria and plasmids are placed into an icey solution, which is then rapidly increased to a hotter temperature, once again allowing the plasmid to penetrate the PM! I think emphasising rapid change is important here as if it were to be gradual then them cholesterol molecules will do their thing and prevent the PM from becoming permeable enough for the plasmids to enter.
So i believe the answer is what you have already mentioned, electroporation i.e the administration of an electrical current, as well as extreme , rapid temperature changes

apologies for yet another question ;/ i've tried searching for this but have failed to find something that sounds reasonable

how do proteins remain stable in low pH environments? (particularly relating to the R side group)

so far i have written 'R side groups on amino acids determine the nature/characteristics of the amino acid. In low pH environments, the R side groups contain features that prevent the protein from denaturing  :-X

any help would be greatly appreciated, thank you! and once again, really sorry to be bothering the bio q thread so much

Well, from my understanding, each enzyme has its own optimum pH - so therefore as you deviate away from this mark, the 3D structure will change. This is because as pH (essentially the measure of H+ ions) gets lower, there are more H+ ions present, which can affect the ionic and hydrogen bonds present.

But, what you've written is good - you'd only really need to specify what the 'features' are, though I can't think of any either ahaha.

Hi all,
At school and also on Edrolo, we were taught about how recombinant plasmids can be created by inserting the desired gene between an antibiotic resistant gene. Then, antibiotic resistance is used to test whether the bacteria has taken up the plasmid and whether the plasmid has taken up the desired gene (is recombinant). I was wondering if this was necessary to know in detail for the course/exam?

Yes, that is something you definitely need to know and has been tested previously. (Refer to Q9 VCAA 2017 Exam)

I'm in some major trouble here friends![/b][/size] I was wondering if anyone has any good ways to remember all that has been taught and all that I've learnt in Biology. I just joined the second semester of VCE Biology Unit 1 and 2 and I was just wondering if anyone had any good tips to help me memorize the content and do well in Biology in the next two years. Also, I was just wondering if help me out with question answering, as I know that in Biology certain terms and phrases must be included, how would I know what to include to get the full marks or if I have written enough for the full marks even after looking at how much a mark is worth?

This thread with some of the best resources that can be useful doing bio.

The powerpoint from the most recent ATARNotes 1/2 bio lecture will give you a good overview of things that you will cover this term.

Douchy's free podcast! (Found here.)

In terms of tips:
I think the biggest thing is to try and deeply understand all information on the study design and how it links together (without learning ridiculous amounts of excess detail in the textbook). Linking concepts and understanding how everything works together allows you to memorise info and answer questions effectively.

As for units 1 and 2 don't worry too much - other than basic biological concepts that come up everywhere it won't be covering most of 3/4 biology which is what actually matters.

I think more recently, biology is less about memorising the content rather the understanding and application of knowledge to certain scenarios and circumstances. Still, you obviously need to know your stuff but straight definition questions are quite a rarity these days. I think everyone is different in terms of memorising - I never actively memorised stuff but tend to remember once I looked over certain stuff multiple times.

For phrases and words required - you definitely need to know a few of these and they are mostly found but completing practice questions/exams so you end up reusing similar answers later on.

Completly agree with sine and Laura here.
I never wrote out any glossaries or worried about specific definitions. I more focused on doing heaps of reading around topics so that I can properly understand them.
Also, you've posted this question here also in the Bio FAQ which is more for the collation of frequently asked questions (as a resource).
For asking questions like the one you have above, its probably best just to ask it here on this thread (just for future reference) :)
